Andy Murray took the court on Sunday looking to secure the point required for Great Britain to defeat the USA and seal a quarter-final berth.
Great Britain were 2-1 up following Friday and Saturday's matches and were counting on the support of Murray's home crowd in Scotland.
Isner started the better of the two in the first set, but galvanised by the support of his fans, Murray managed to fight his way to a tie break and a 7-6 win in the first set.
The Scot stepped up a gear in the second set and broker Isner's serve in the sixth game with a brilliant lob.
Murray eventually securing the set 6-3.
Despite the routine win in the second set, the world number five was made to work hard in the third with Isner refusing to give in.
Murray requiring another tie break to seal the deal for a 7-6 6-3 7-6 win.
Great Britain take an unassailable 3-1 lead in the tie and set up a home quarter-final with France in July.
This after the French won qualification over Germany in their tie having taken an unassailable 3-0 lead on Saturday.
The match ended 3-2 after Philipp Kohlschreiber and Jens-Lennard Struff both won their games on Sunday to ensure some pride was restored.
