His compatriot Rui Machado had shocked Dmitry Tursunov earlier in the day and Sousa seemed inspired as he won the first set 6-3.
But Sousa faltered in the second as Mayer took the match to a deciding set.
The momentum was with Argentina's Mayer.
And he wrapped up the 3-6, 6-1, 6-2 victory to set up a second round tie with Kazhakstan's Andrey Golubev.
