Queen's Park host Albion Rovers with Jamie Brough, Paul Harkins, Steven Baillie, Michael O'Hara and David Murray all unavailable through injury.
The Spiders reached the Play-offs last season, but they fell at the first hurdle after losing 4-0 to Arbroath in the first leg and Speirs is determined that there will be no repeat.
Head coach Gardner Speirs said: "Last year did not go so well but this is a different occasion with different personnel and we will be looking to make more of an impact.
"After a disappointing start to the league season we have done well to turn things around but we do not want to stop now."
Albion Rovers have been dealt a blow ahead of their trip to Queen's Park after on-loan Motherwell striker Steven Lawless was ruled out with a thigh strain.
Manager Paul Martin is still waiting on news of Steven Canning (hand injury) and Chris Boyle (hamstring).
Yet striker John Gemmell is fit enough to be considered for a starting role after several substitute appearances.
Martin said: "Not having Steven Lawless available is a disappointment but we will just have to get on with things.
"We are excited to be in the play-offs for the first time and determined to make sure we do the best that we can.
"Two-legged games are new to us but we are looking to have another two-legged game next week."
